Electron Microscope Autoradiographic Identification of Dopamine Synthesizing APUD Cells in the Rabbit Gastrointestinal Mucosa*

Abstract
Electron microscopic autoraidographic observations on the rabbit gastrointestinal APUD cell series were made in order to demonstrate the intracellulardistribution of 3H-dopamine after an injection of 3H-dopa as a precursor.
One hour after an injection, the pieces of rabbit gastrointestinal mucosa wereremoved and fixed in a 2.5% glutaraldehyde solution in 0.1M cacodylate buffer(pH 7.4) and postfixed in 1.5% OsO4 in 0.3M cacodylate buffer (PH 7.4). Tissueswere then dehydrated and were embedded in epoxy resin. Ultrathin sections were affixed on a 2 % celloidin coated glass slide, covered with a monolayer of ILF ORD L4 emulsion by dipping method and then exposed at 4¡ÆC in a light-tight. box for two months and then developed in microdol X and fixed in 30% sodium hyposulfide.
Several cell types of APUD series were clearly identified in the rabbit gastrointestinal mucosa. Of them D cells in fundus, G cells in pylorus and the cell types with high pleomorphic secretory granules¢¥ in colon were shown high radioactivity in their cytoplasm, but radioactivity in APUD series of duodenal mucosa were relatively low.
Based on the above results, it is concluded that dopamine synthetic activities from APUD cell series were variable according to the types of APUD cell.
